To investigate the role of AHb1 in plants we characterized the interaction of AHb1 with NO by biochemical and genetic approaches. Here we provide evidence to suggest that AHb1 functions to metabolize NO to nitrate utilizing NADPH. AHb1 interacts with NO in vivo to form Snitroso hemoglobin and reduces the NR-dependent NO emission during transfer of lightadapted plants to darkness and under hypoxic stress. However, AHb1 does not interfere with NO burst originated by acute responses, and it does not attenuate hypersensitive cell death, when NO-mediated signaling functions are needed.
