Mg- and K-ATPase activity was detected in the microsomal fraction from roots of grapevine (Vitis vinifera L. cv. Verduzzo) seedlings. The presence of several additives in the extraction medium, such as choline-Cl, ethanolamine, glycerol-1-P, EDTA, BSA, PVPP and DTT was necessary for obtaining active enzyme preparations. Optimum pH for enzyme activity was 6.5 in the presence of both Mg2+ and K+. Enzyme activity was inhibited by vanadate and DES but not by molybdate and scarcely by azide.
